ADAR
ADAR ,全稱為腺苷脫氨酶作用于RNA 1,是RNA編輯的關鍵酶,能夠將RNA分子中的腺苷(A)轉化為肌苷(I),從而影響RNA的功能和編碼的蛋白質。ADAR 蛋白在多種生物學過程中發揮重要作用,包括免疫反應、神經功能和疾病的發生發展。ADAR 蛋白相關疾病包括Aicardi-Goutières綜合征(AGS)和遺傳性對稱性色素異常癥(DSH),這些疾病都與RNA編輯異常有關。目前,針對ADAR蛋白的藥物研發主要集中在RNA編輯療法,旨在通過精確修飾RNA序列來治療遺傳疾病和復雜疾病。例如,斯坦福大學生物學家李進聯合創辦的RNA編輯療法公司AIRNA,正在開發針對Alpha-1抗胰蛋白酶缺乏癥(AATD)的RNA編輯療法,計劃于2025年進行I期臨床試驗。此外,其他公司如Wave Life Sciences和Korro Bio也在積極開發基于RNA編輯技術的藥物,為治療AATD等疾病帶來新的希望。

IHC image of CSB-RA906343A0HU diluted at 1:100 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
