    The gene encoding the 25-300aa of the rabbit TFPI was fused with the 10xHis-tag gene to form a synthesized gene, which was cloned into a plasmid vector. The vector clone was transfected into a mammalian cell line for subsequent expression. The cell culture supernatant was purified to obtain the recombinant rabbit TFPI protein. The purity of this recombinant TFPI protein is greater than 95%, measured by SDS-PAGE. The LAL method evaluates its endotoxin level as less than 1.0 EU/ug. It has also been validated to be an active protein. In the functional ELISA, this recombinant rabbit TFPI protein can bind to the recombinant antibody against TFPI, with the EC50 of 2.281-3.783 ng/mL.

    TFPI is an endogenous plasma anticoagulant protein and the natural inhibitor of the tissue factor (TF)-mediated coagulation pathway. TFPI binds to and inactivates FXa and then quickly combines with the TF/FVIIa complex, forming the quaternary complex FXa-TFPI-T-FVIIa that blocks the activation of the coagulation cascade. Rabbit TFPI protein has potential clinical application prospects as it has been used in the process of the preparation or screening of antibodies against TFPI. Therefore, developing TFPI proteins of rabbits and other related species can rapidly promote numerous TFPI antibody drugs into the clinic.
