    CUSABIO produced the polyclonal antibody against lktA by immunizing rabbits with a synthetic peptide that corresponds to amino acids 715-953 of the recombinant Leukotoxin protein. The antiserum obtained from the rabbits was purified using protein G affinity chromatography, resulting in a purity level of 95% for the anti-lktA antibody.

    This antibody is an unconjugated IgG that specifically reacts with Escherichia coli lktA. LktA is a virulence factor generated by the bacterium Mannheimia haemolytica, which targets and kills immune cells, particularly leukocytes, in the lungs of infected animals. The lktA polyclonal antibody developed by CUSABIO is suitable for detecting the lktA protein in ELISA.

    Pasteurella leukotoxins are exotoxins that attack host leukocytes and especially polymorphonuclear cells, by causing cell rupture. The leukotoxin binds to the host LFA-1 integrin and induces a signaling cascade leading to many biological effects, including tyrosine phosphorylation of the CD18 tail, elevation of the intracellular Ca(2+) and lysis of the host cell. This leukotoxin is a major contributor to the pathogenesis of lung injury in ovine pneumonic pasteurellosis. It has also weak hemolytic activity.
