TET2
TET2,甲基胞嘧啶雙加氧酶2,別名Ten-eleventranslocation2(TET-2)。它是一種α-酮戊二酸和Fe2+依賴的雙加氧酶,主要來源于哺乳動物的TET蛋白家族,該家族包括TET1、TET2和TET3三個成員。TET2的作用機制是通過催化5-甲基胞嘧啶(5mC)氧化生成5-羥甲基胞嘧啶(5hmC),進而通過一系列的氧化步驟最終轉化為未修飾的胞嘧啶,這一過程是DNA主動去甲基化的重要途徑。TET2在細胞命運決定、分化、腫瘤發生發展以及免疫調控中發揮重要作用。TET2的異常表達或突變與多種疾病相關,包括髓系腫瘤和免疫相關疾病。在腫瘤抑制方面,TET2通過調控基因表達和DNA甲基化狀態,影響細胞增殖和分化。在免疫調控中,TET2能夠影響炎癥因子的分泌,參與炎癥反應的調控。
