P2RX3
P2RX3,即嘌呤能受體P2X配體門控離子通道3型蛋白,常被稱為P2X3受體,別名P2X3。P2RX3屬于ATP門控的非選擇性陽離子通道家族,主要分布于感覺神經元末梢,通過與胞外ATP結合觸發鈉、鈣離子內流,參與痛覺信號傳導及內臟器官的神經反射調控。研究發現,其過度激活與慢性咳嗽、膀胱過度活動癥及糖尿病神經病變密切相關,在神經源性炎癥和內臟超敏反應中起關鍵作用。近年來針對P2X3受體的藥物開發取得突破,如首個高選擇性拮抗劑Gefapixant(AF-219)已完成Ⅲ期臨床試驗,證實能顯著改善難治性慢性咳嗽患者癥狀,但存在味覺障礙副作用。另有Eliapixant(BAY-1817080)等二代藥物通過結構優化降低不良反應,目前處于Ⅱ期研究階段。該靶點同時被探索用于治療間質性膀胱炎和神經病理性疼痛,其雙重拮抗劑(同時阻斷P2X2/3異源三聚體)在腸易激綜合征等疾病中展現出潛在治療價值。
