KIF11
KIF11(驅動蛋白家族成員11)又稱Eg5或Kinesin-5,是一種微管相關馬達蛋白,通過水解ATP產(chǎn)生機械力推動微管滑動。它在有絲分裂中驅動雙極紡錘體的形成與分離,確保染色體精確分配到子細胞,若功能異常可能導致染色體非整倍性,進而引發(fā)細胞癌變或凋亡。該蛋白異常高表達常見于乳腺癌、結直腸癌等實體瘤,與腫瘤增殖、轉移及不良預后相關。此外,KIF11基因突變可導致常染色體顯性小頭畸形、視網(wǎng)膜變性等罕見神經(jīng)發(fā)育疾病。
針對KIF11的抗癌藥物研發(fā)聚焦于抑制其ATP酶活性,代表性化合物如ispinesib、filanesib通過阻斷紡錘體分離迫使癌細胞停滯在分裂中期。盡管早期臨床試驗在血液腫瘤中顯現(xiàn)療效,但實體瘤響應有限且存在神經(jīng)毒性問題。近年研究轉向聯(lián)合用藥策略,如Eg5抑制劑與紫杉醇聯(lián)用可增強抗微管藥物敏感性。新型變構抑制劑ARRY-520已完成II期試驗,而PROTAC技術開發(fā)的降解劑正探索克服耐藥性的可能。
