GALR3
甘丙肽受體3(GALR3)是一種G蛋白偶聯(lián)受體,屬于甘丙肽受體家族,與神經(jīng)肽甘丙肽結(jié)合。該受體廣泛分布于中樞和外周神經(jīng)系統(tǒng),在海馬、下丘腦、杏仁核等區(qū)域表達(dá)量較高。GALR3的激活能夠調(diào)節(jié)多種生理過程,包括認(rèn)知、情緒、痛覺、進(jìn)食、神經(jīng)內(nèi)分泌和能量代謝等。在抑郁癥中,GALR3與GalR1和GalR3共同介導(dǎo)抑郁癥狀,而GalR2則發(fā)揮抗抑郁作用。GALR3的激活會(huì)抑制腺苷酸環(huán)化酶,打開內(nèi)向整流鉀離子通道,并激活絲裂原活化蛋白激酶。此外,GALR3還與多種神經(jīng)遞質(zhì)和神經(jīng)肽相互作用,如去甲腎上腺素、5-羥色胺、多巴胺、神經(jīng)肽Y等,共同調(diào)節(jié)情緒、痛覺和進(jìn)食行為。GALR3相關(guān)疾病包括癲癇、酒精中毒、神經(jīng)病理性疼痛、糖尿病和癌癥等。目前,針對(duì)GALR3研發(fā)的藥物主要集中于抗抑郁和抗焦慮領(lǐng)域,如GalR3拮抗劑SNAP 37889和選擇性GalR2激動(dòng)劑AR-M1896等,這些藥物在動(dòng)物實(shí)驗(yàn)中顯示出一定的療效,但仍需進(jìn)一步研究以評(píng)估其安全性和有效性。
