E7蛋白具有轉化活性和反式激活活性。 結構為兩個功能區域:一個是N末端區域,含有保守的Cys-X-X-Cys(CXXC)結構,用于與細胞中的特定蛋白質相互作用;另一個是C末端區域,包含保守的LXCXE序列,用于與細胞中的Rb蛋白(Retinoblastoma protein)結合。E7蛋白通過與細胞中的Rb蛋白結合,抑制Rb蛋白的功能,導致細胞周期的失控和異常增殖。這會使細胞失去正常的生長抑制機制,促進細胞轉變和癌變。E7靶點及其與Rb蛋白相互作用的研究已成為HPV相關癌癥治療和疫苗研發的重要方向,是HPV感染中的關鍵蛋白之一,它與細胞周期調控、轉錄調控和細胞增殖等過程密切相關。
